Prior hydrolysis, the Rhodamine110 moiety is quenched resulting in a very low background.

This substrate is ideal for activity determination of 3CLpro and suitable for HTS or pilot screens in search for potential inhibitors due to its red shifted fluorescence compared to AMC or Dabcyl/Edans substrates. This results in less interference with potential inhibitors in screeing campaigns.
