UCHL3 is a deubiquitinating enzyme (DUB) that controls levels of cellular ubiquitin through processing of ubiquitin precursors and ubiquitinated proteins. It is a thiol protease that recognizes and hydrolyzes a peptide bond at the C-terminal glycine of either ubiquitin or NEDD8. It digests precursors and ubiquitinated proteins to generate monomeric ubiquitin. This UCHL3 is recombinatly expressed in E.coli.[1][2][3][4][5]

Purified recombinant human Sentrin Specific Protease 1 (SENP1, catalytic domain) is a SUMO-specific enzyme that catalyzes two essential functions. Processing the full-length versions of SUMO1, SUMO2, and SUMO3 into its mature forms and the deconjugation of SUMOylated proteins. This SENP1 is N-terminally His6 tagged and expressed in E.coli.[1][2][3][4]

The severe acute respiratory syndrome coronavirus papain-like protease (SARS-CoV His6-PLpro) is involved in the processing of the viral polyprotein. Proteolytic processing of the coronavirus replicase poly-protein is essential for generating a functional virus replication complex. PLpro is able to hydrolyze both ISG15-Rhodamine110 or diubiquitin/tetra-ubiquitin substrates, but is very inefficient when processing mono-Ub conjugates or synthetic peptide substrates. This PLpro is N-terminally His6 tagged and expressed in E.coli. [1][2][3][4]

NEDP1 is a protease which hydrolyzes the isopeptide bond between the C-terminus of NEDD8 and the respective lysine of the substrate protein to which the NEDD8 is covalently bound. This NEDP1 is his-tagged on the N-terminus, recombinantly expressed in E. coli.[1][2]

The human SARS-CoV-2 coronavirus harbors two proteases Papain Like Protease PLpro and 3CLpro (Chymotrypsin like Protease) or Mpro (Main-Protease) which is a C30-type cysteine protease. The viral genome encodes more than 20 proteins, with 3CLpro located within the non-structural protein 5 (nsp5) section of the viral polypeptide that cleaves together with PLpro polyproteins (PP1A and PP1AB) into individual functional components.

The severe acute respiratory syndrome coronavirus papain-like protease (SARS-CoV-2 PLpro) is involved in the processing of the viral polyprotein. Proteolytic processing of the coronavirus replicase poly-protein is essential for generating a functional virus replication complex. PLpro possesses both deubiquitinating or deISGylating activity and can process Lys- 48 and Lys-63 linked polyubiquitin chains (free chains or from cellular substrates).
